Oh. I'm sorry, it appears I had confused the Addrbook file (which I don't use) with .mew-alias. The latter can be edited by hitting M-a in Summary mode, and the contents then get automagically loaded when the file is saved.

There does not seem to be an equivalent mechanism for the Addrbook file. From poking around in mew-addrbook.el, I guess that running mew-addrbook-setup will do what you want, but since that function is not made interactive and seems intended to only be run on startup, I cannot guarantee that running it on your own will not have undesired side effects.
